Zoë Yadira Saldana est née le 19 juin 1978 à Passaic NJ d'une mère portoricaine et d'un père dominicain. Sa famille a déménagé en République dominicaine à l'âge de dix ans. Là, elle a pratiqué le ballet dans l'une des écoles de danse les plus prestigieuses du pays. Saldana est retournée à New York à l'âge de 17 ans, où elle a commencé à s'impliquer dans des groupes de théâtre tels que Faces et le New York Youth Theatre. Sa formation de ballet l'a aidée à débarquer sa première partie à l'écran en tant qu'Eva dans le film de danse, Centre Stage (2000). La carrière de l'actrice a continué à prospérer avec des rôles dans Pirates des Caraïbes: The Curse of the Black Pearl (2003) avec Johnny Depp, The Terminal (2004) avec Tom Hanks, et Guess Who (2005) face à Ashton Kutcher. Saldana continue d'agir et de modéliser avec des fonctionnalités éditoriales dans Elle, Vanity Fair, V, GQ Italia et Nylon.
